    IIS server (Windows 2008 R2, IIS 7) connecting to the same SQL Server database. I'm now getting an error 500 which means that I can't connect to the database, error:  login failed for user 'IIS APPPOOL\ASP.NET v4.0', tried adjusting permissions to the database which further screwed things up. Any ideas how to configure IIS APPPOOL\ASP.NET v4.0 to get round this issue.

    Run the application pool under a domain user and then assign that user account permissions on the SQL Server database please. You are not supposed to use "IIS APPPOOL\ASP.NET v4.0" when you have no idea what it is.
