    If excel does not exist, you can create it by office directly or use Open XML SDK, then insert excel data into Sql Server. In this thread Yang Shen had answered your question.

    Do you mean that this excel is in opening status? 

    If excel is open, then it is not possible to read and write to it in asp.net.
