    No, Windows 2012 R2 doesn't support installation of Biztalk 2009. Please refer below article for more reference,

    Officially BizTalk 2009 is not supported on Windows 2008 R2 too.

    Please refer below thread for reference,

    Though,

    Announced at PDC 2009, Biztalk 2009 R2 will additionally support:

    •  Windows Server 2008 R2

  • BizTalk Server 2009 will run on Windows Server 2008 R2 but it is not a supported configuration.  You can get away with it for most DEV use but your Build and Production servers must be a supported configuration.

    Altough this is doable and the system works you wont be able to get support from Microsoft if something fails due to the non supportive setup and configuration. Please refer to the reference added by Kamlesh Kumar (https://support.microsoft.com/en-us/kb/926628 )
