Some users are not able to receive alerts form library although previously they were able to do so. Any suggestion ?

Some or all?  If all, then check the Outgoing email settings in Central Administration.  Make sure there isn't an antivirus installed locally on the SharePoint Server that is blocking outgoing email (or tcp/25) as well as validate that the settings are correct.  Also make sure the SMTP server does not require authentication or that it doesn't have an allowed-by IP list, etc.

As i said only few users are not able to receive alerts, I have also tried to Add alert(form problematic users system) for specific library but it gives Access denied error.

I did it after setting alert from sharepoint admin. It works like a charm. Thanks for your support.

Recheck your alert option again tehre are option to send you alert if you make any changes.. select it .. then there is one more option send alert immediately .. set that one either.  Then see if you get alert.

I Checked the Web application general setting:

Alerts on this Server are: On

Maximum number of alerts that a user can create:  Unlimited number

    So alert still now working.
