Thanks for your post.
In order to resolve this issue I want to confirm some information with you:
1 Could you please provide the detailed information about your TFS configure environment?
2 Have you check the TFS Server machine, the CPU and Memory on TFS Server machine are enough to use at that time?
3 Open a small size file under Source Control Explorer or only query several work item in a Query still response very slow?
4 If you installed VS 2010 on the TFS 2010 Server machine, try to open file and query work item on the TFS 2010 Server machine, still slow or not?
5 Your TFS 2010 response is very slow before? Or after installed something, it became slow?
6 Have you tested this scenario on other clients, still slow?
And I suggest you upgradeyour TFS 2010 and VS 2010 both to TFS 2010 SP1 and VS 2010 SP1.
And try to clean the Cache manually:

- Clean the Cache folder on Server machine. The folder path is:
C:\Users\username(Web access user name)\AppData\Local\Microsoft\Team Foundation\3.0\Cache(os: Windows Server 2008 R2)

What type of infrastructure do you have supporting your deployment?
And if you can provide the info John Qiao suggested, that would give more background on your situation.
